Adapting Ristocetin Cofactor Assay Reagent

Has anyone been able to adapt a third-party reagent for ristocetin onto a Stago STAR-E or Compact? If so, would you be willing to share your experience with the product, validation procedure, etc? Please feel free to reply off-line if you so desire, and thank you in advance,

Hello, Dorothy, and thank you for your question. I’ve known of one facility that successfully adapted the Siemens automated ristocetin cofactor assay reagent on a Stago STA several years ago, and another that was unable to use the reagent on the STAR-T. I’ll be interested in learning if any of our subscribers have made this work. Geo

From Dave McGlasson: Dorothy asked about the ristocetin co-factor (RCF) assay on the STA-R Evolution. I would suggest forget it and use the Beckman/IL LIA VWF assay. It worked great on our Evolution and have used it in a couple of protocols for publications. The RCF assay uses the Siemens formalinized platelets. We accomplished this procedure several years ago but it was cumbersome with CVs > 10%. The Beckman IL VWF LIA test had CVs < 5% on between-run and within-run testing for precision. It also had great sensitivity and specificity characteristics. Stago and Beckman/IL have test set-ups for the LIA method.

We were able to adapt the Siemens automated ristocetin assay on our Stago STA-R Evolution. Linearity down to 0.10 IU/mL. We presented a poster at ISTH this past year. I can email you with our test set-ups if you’re still interested.

We were able to adapt the Beckman IL vWF LIA test on the STA Evolution at Baptist Medical Center in San Antonio, TX. Stago has the test set ups. We use Precision BioLogic Normal Ref for the calibrator.
